So far, turnout around the country is reported to be low, with many places reporting just a "trickle" of voters.
Supporters of the proposed amendment say it will strengthen child protection legislation, but some opponents of the measure claim it will hand too much power to Government agencies.
A result is expected some time tomorrow afternoon.
Today is only the second time a referendum has been held on a Saturday. The first was the second vote on the Nice Treaty a decade ago.
The experiment was aimed at boosting turnout, but initial interest seems to be relatively low.
The lowest ever turnout was in July 1979, when 28.5% of eligible voters turned out to cast their ballots on the issues of adoption and university representation in the Seanad.
Tánaiste Eamon Gilmore said last week that future weekend polls hinged on the turnout figure today.
It has also been claimed that the later than usual start to voting will save over €500,000.
Voters are urged to bring identification as well as their polling card to the polling station. If voters do not have official notification to hand, ID such as a driving licence or a passport will suffice.
The Chairperson of the Referendum Commission, Ms Justice Mary Finlay Geoghegan, has urged voters to try to find the time to cast their ballots today.
